The prodrug mechanism. THCo acetate is pharmacologically inactive in its native form. It does not meaningfully bind CB1 or CB2 receptors until the acetate group is cleaved off. That cleavage happens after ingestion, primarily in the liver, via carboxylesterase enzymes. Once deacetylated, the molecule becomes Delta-9 THC, which then distributes into the bloodstream, crosses the blood–brain barrier, and activates the standard endocannabinoid cascade — CB1 partial agonism in the central nervous system, CB2 binding in immune and peripheral tissues, and downstream modulation of dopamine, serotonin, GABA, and glutamate signaling. Translation: this is still THC at the receptor, but more of it gets there, faster, and at a higher peak.

Pharmacokinetics by route. Oral ingestion produces the most dramatic and longest-lasting effects because of first-pass hepatic activation: onset typically 30–60 minutes, peak 2–3 hours, total duration 6–10 hours, sometimes longer at higher doses. Sublingual (held under the tongue, then swallowed) produces a faster onset, similar to oral but with somewhat earlier peak because a fraction absorbs directly through the oral mucosa. The metabolite you eventually make from THCo is the same metabolite you make from any THC — 11-hydroxy-THC (11-OH-THC) for oral, plain Delta-9-THC for inhaled — which is why standard THC drug interactions, drug testing, and impairment timelines all apply.

⚠️ The Safety Science You Must Read Before You Dose

We are about to repeat this in three different places on this page because it is the single most important thing you can know about this product: THCo acetate is not safe to vaporize at high temperatures. Research published in 2023 by a team at the University of Michigan (J. Chromatogr A / Drug Test Anal) found that when THCo acetate is heated to approximately 340°C (644°F) — the upper end of what many unregulated vaporizer devices and dab rigs can reach — it can degrade and release ketene gas, a toxic inhalant that is potentially lethal at concentrations as low as 5 ppm. Ketene is a documented pulmonary irritant and is one of the compounds implicated in the 2019 EVALI (e-cigarette or vaping product use-associated lung injury) outbreak that sickened thousands of Americans and killed dozens. It is also the reason vitamin E acetate was banned in regulated vape cartridges — the same chemistry, the same warning.

What this means for how you use this product. If you choose to inhale, use the lowest effective temperature on a device you can verify — ideally a quality concentrate vaporizer with a precise temperature display, set below 315°C (600°F), and never let the chamber glow visibly. Prefer sublingual or oral administration whenever possible — the deacetylation happens in your liver either way, and you entirely sidestep the pyrolysis question. Never use this distillate in a cheap pen, a sub-ohm mod, or any device whose actual coil temperature you cannot confirm. If you are an experienced daber with a temperature-controlled e-rig and you know what you are doing, fine — but if you have to ask, the answer is sublingual.

Is THCo the same as Delta-9 THC?

No. THCo (THCo acetate) is a semi-synthetic derivative that the liver has to convert into Delta-9 THC before it activates the CB1 and CB2 receptors. The end metabolite is the same, but the prodrug curve (delayed onset, higher peak, longer duration) and the potency are meaningfully different. If you are comfortable with 5–10 mg of edible Delta-9, do not start with 5–10 mg of THCo.

Will THCo show up on a drug test?

Yes. After hepatic conversion, the metabolites in your system are identical to those produced by any other THC ingestion — primarily 11-OH-THC and THC-COOH.
